    Applications

    2-Mercaptoethanol is used to reduce disulfide linkages in solubilizing proteins for gel electrophoresis (typically used in SDS-PAGE sample buffer at 5% concentration). Also reduces excess oxidative polymerization of catalysts. Cleaving intermolecular (between subunits) disulfide bonds allows the subunits of a protein to separate independently on SDS-PAGE. Cleaving intramolecular (within subunit) disulfide bonds allows the subunits to become completely denatured so that each peptide migrates according to its chain length with no influence due to secondary structure. In solution, 2-mercaptoethanol is readily oxidized in air to a disulfide, especially at alkaline pH. Because of this property, it is widely used to protect proteins, enzymes in particular, from becoming inactive. An excess of 2-mercaptoethanol (generally used at 0.01 M) will maintain the protein thiol groups in their reduced state.

    Applications

    Brilliant Blue G is a useful dye for SDS gels as it readily stains proteins with minimal background color. Protein bands can be visualized during staining. It has been used in the Bradford dye-binding protein assay. A mechanism for dye binding to protein has been proposed, based onmeasuring Coomassie Brilliant Blue G-250 (CBBG) absorbance spectra during titration of the dye reagent in the absence of protein and its response to different polyamino acids. Brilliant Blue G has been used for determining critical micelle concentration of detergents. Recent research reveals CBBG as a P2X7 (P2X purinoceptor 7: P2RX7) antagonist and suggests that it exhibits neuroprotective properties in rat spinal cord cells.

    Applications

    Ethidium bromide is the most commonly used nucleic acid stain for PAGE or agarose gel electrophoresis. The fluorescence of EtBr increases 21-fold upon binding to double-stranded RNA and 25-fold on binding double-stranded DNA so that destaining the background is not necessary with a low stain concentration (10 μg/mL). Ethidium bromide has been used in a number of fluorimetric assays for nucleic acids. It has been shown to bind to single-stranded DNA (although not as strongly) and triple-stranded DNA. Because of its ability to bind to DNA, EtBr is an inhibitor of DNA polymerase.

    Quercetin hydrate was used as leishmanicidal flavonol to investigate the inhibition of recombinant arginase from Leishmania amazonensis by quercetin, quercitrin and isoquercitrin.[1]

    General description

    Quercetin is an antitrypanosomal and antileishmanial compound.[1] Quercetin is a well-known bioflavonoid and its influence on tumour growth in vivo has been reported.[2] Quercetic exhibits leishmanicidal effect on the amastigote stage of Leishmania donovani.[1]
